Ly49receptorsinrodents,likeKIRsinhumans,regulateNKcellactivity.AlthoughinhibitoryLy49receptorsclearlyrecognizeMHC‐Imolecules,ligandsfortheactivatingLy49receptorsarelesswelldefined.Here,weshowthattheactivatingLy49s4andtheinhibitoryLy49i4receptorsrecognizenonclassicalMHC‐ImoleculesontheratmacrophagecelllineR2(RT1d).ListeriainfectionofR2macrophagesledtoincreasedexpressionofclassicalandnonclassicalMHC‐Imolecules.CoincubationoftheseinfectedcellswithreportercellsexpressingLy49i4orLy49s4increasedthereportercellresponses.TheseresponseswereblockedbymAbOX18(anti‐MHC‐I)andAAS1(anti‐nonclassicalMHC‐I).IFN‐γtreatmentofnormalR2cellsalsoincreasedtheMHC‐Iexpressionandenhancedthereportercellresponses.TheseresultssuggestthatactivatingandinhibitoryLy49receptorsmonitorMHC‐IexpressiononListeria‐infectedcells...
