Thisstudyaimedtoidentifytheinflammation‐associated7/4‐antigen,whichishighlyexpressedonneutrophils,inflammatorymonocytes,someactivatedmacrophages,aswellasonbonemarrowmyeloid‐restrictedprogenitors.Thehighexpressiononinflammatorycellsissuggestiveofaroleininflammationandmakesthe7/4‐antigenapotentialtargetforthemanipulationofinflammatorycells.Consistentwiththis,the7/4‐antibodymediatesspecificdepletionof7/4‐expressingneutrophilsandmonocytes.Wehaveidentifiedthe7/4‐antigenasa25‐to30‐kDaGPI‐anchoredglycoproteinsynonymouswiththeLy‐6B.2alloantigen.WecharacterizedtheexpressionofLy‐6Bduringtheinflammatoryreactioninducedbyzymosan.Duringthelaterstagesofanexperimental,acute,self‐resolvinginflammatoryresponse,wefoundthatLy‐6Bisdifferentiallyexpressedonmacrophages.Ly‐6B‐expressingmacrophagesalsoexpressmoreMHCII,CIITA,CCR2,Ly‐6C,andCD62LthantheLy‐6B‐negativemacrophages,whichinturn,expressmoreoftheresidenttissuemacrophagemarkerSIGN‐R1andhigherCD11bandF4/80.Ly‐6B‐expressingmacrophagesincorporatemoreBrdUthantheirLy‐6B‐negativecontemporarieswhenfedduringtheresolutionphaseoftheacuteinflammatoryresponse.Thus,Ly‐6Bexpressiononmaturemacrophagesdefinesasubsetofrecentlygeneratedinflammatorymacrophagesthatretainmonocyticmarkersandishenceasurrogatemarkerofmacrophageturnoverininflammatorylesions.Thedefinitionofthe7/4:Ly‐6BantigenwillallowfurthercharacterizationandspecificmodulationofLy‐6B‐expressingcellsinvivo...
