Neutrophilsaresentinelcellsthatexpressinhighervertebrates>30chemokineandchemoattractantreceptorstosenseandquicklyreacttotissuedamagesignals.Intravitalmicroscopystudiesinmousemodelsofwounding,inflammation,andinfectionhaverevealedthatneutrophilsformcellswarmsatlocalsitesoftissueinjuryandcelldeath.Thisswarmingresponseischoreographedbychemokines,lipids,andotherchemoattractants,controllingsequentialphasesofhighlycoordinatedchemotaxis,intercellularsignalrelay,andclusterformationamongneutrophils.Thisreviewwillgiveabriefoverviewaboutthebasicprinciplesandkeymoleculesthathaveledtotherefinedmultistepmodelofhowneutrophilscometogethertoisolatesitesoftissueinjuryandmicrobialinvasionfromhealthytissue.Whereasauto‐andparacrinesignalingamongneutrophilsduringlaterphasesofswarmingcanprovidealevelofself‐organizationforrobustnavigationindiverseinflammatorysettings,guidancefactorsfromprimarytissuelesions,residentbystandercells,anddyingcellsregulatetheinitialphasesoftheswarmingresponse.Thisreviewwilldiscusshowthespecificenvironmentalcontextandmixtureofattractantsatthelocallyinflamedsitecanleadtovariantsofthemultistepattractionmodelandinfluencetheextentofneutrophilswarming,rangingfromaccumulationsofonlyfewindividualcellstotheaggregationofseveralhundredsofneutrophils,asfoundinabscesses.Giventhecriticalrolesofneutrophilsinbothhostprotectionandtissuedestruction,novelinsightsonneutrophilswarmingmightprovideusefulforthetherapeuticmodulationofneutrophil‐dependentinflammatoryprocesses...
