DuringMS,phagocytosingmyelin‐containingmacrophagesariseandlieincloseproximitytoTcells.Todate,ithasnotbeenaddressedwhetherthesemyelin‐ladenmacrophageshavethecapacitytopresentantigenstoTcellsandwhetherthiscontributestoinflammationindisease.Wedemonstratethatinvitro‐generatedhumanandmousemyelin‐ladenmacrophagesexpressedMHCclassIandIIandcostimulatorymoleculesandarethuswellequippedforantigenpresentation.Humanmyelin‐ladenmacrophagesexhibitednormalendocytosisofparticulateandsolubleantigens.Inaddition,humanmyelin‐ladenmacrophageselicitedactiveTcellproliferationofnaïveaswellasmemoryTcells.Furthermore,mousemyelin‐ladenmacrophagesinducedprimaryantigen‐specificCD4+TcellproliferationinvivobuttransientlydiminishedIFN‐γrelease.Functionally,MOGpeptide‐loadedmyelin‐ladenmousemacrophagesmodestlybutsignificantlyreducedtheseverityofMOGpeptide‐inducedEAE.Thesedatashowthatmyelinuptakeresultsintheinductionofapopulationofmacrophagesthatretainsantigen‐presentingcapacityandlimitsautoimmune‐mediateddisease...
