SOCEviaCRACchannelsisacriticalsignalingeventinimmunecells.Recentstudieshaveidentifiedkeyproteinsunderlyingthisprocess;STIMisanERCa2+sensorthatinteractswithOrai,anintrinsic,pore‐formingproteinoftheCRACchannel.Inheterologousexpressionsystems,STIM1regulatesSOCEbyinteractingwithOrai1,‐2,and‐3.Innativetissues,however,thepreciserolesofSTIMandOraiproteinsarenotwelldefined.Here,wehaveinvestigatedthemolecularcomponentsofSOCEsignalinginmouseDCs.WeshowthatDCspredominantlyexpressSTIM2andonlyverylowlevelsofSTIM1comparedwithTlymphocytes.UponstoredepletionwithTg,STIM2aggregatesandinteractsselectivelywithOrai2.Incontrast,TgfailstoaggregateSTIM1orenhanceSTIM1‐mediatedinteractionswithOraiproteins.Consistentwiththisbiochemicalcharacterization,stimulationofDCswiththeadhesionmoleculeICAM‐1selectivelyrecruitsSTIM2andOrai2totheIS.Together,thesedatademonstrateanovel,STIM2‐dependentSOCEsignalingpathwayinDCs...
