【 摘 要 】
Th1andTh17cellshavebeenconsideredaseffectorsinmouseEAEandinthehumancounterpart,MS.Recently,IL‐22,aTh17‐related,proinflammatorycytokine,hasbeenassociatedwithanewThcellsubset,definedasTh22,involvedinchronicinflammatoryconditions,suchaspsoriasis;theroleofIL‐22inMShasnotyetbeenelucidated.Here,wereportthatsimilartoTh17cells,thenumberofTh22cellsincreasedinthePBandtheCSFofRRMSpatients,especiallyduringtheactivephasesofthedisease.However,asopposedtoTh17cells,theexpansionofTh22cellsoccurredbeforetheactivephasesofthedisease.Th22cellswerefoundtobespecificfortheautoantigenMBPandalsoexpressedhighlevelsofCCR6andT‐bet,asforTh17cells,indicatingthatTh22self‐reactivecellscouldhaveCNS‐homingpropertiesandbepathogenicinactiveRRMSpatients.ConverselytoTh17cells,Th22cellsdisplayedlowerlevelsofIFNAR1andwereinsensitivetoIFN‐βinhibition.ThesedatasuggestthatexpansionofTh22cellsinMScouldbeoneofthefactorsthatcriticallyinfluenceresistancetoIFN‐βtherapy...
