Thestromalmilieu(cellularandmatrixcomponents)helpsestablishtissue“address‐codes”thatdirectleukocytebehaviorininflamedtissue.Coordinatedinteractionsamongthestroma,leukocytes,andECsdictatewhichleukocytesarerecruited,whethertheyareretainedwithintheinflamedsite,andhowlongtheysurvive.Herein,wediscusshowthestromalmilieuinfluencestheleukocyterecruitmentcascade.Moreover,weexplorehowcorruptionofthestromalphenotypeinchronicinflammatorydiseasescontributestoundesired,continuousrecruitmentofleukocytes.Emergingcomplex,multicellular,multilayered(co‐)culturemodelsarenowaddressingthemolecularcircuitryinvolvedinregulatingstromalorganizationduringinflammation.Understandingcontext‐specificchangesinpro‐oranti‐inflammatoryagentsderivedfromthestroma,suchasIL‐6(anditscofactors),isimportantforthegenerationoftherapeuticstrategiesthatrestorethebalancebetweenrecruitmentandclearanceoftheinflammatoryinfiltrateinchronicdisease...
