UseofSPR‐basedbiosensorsisanestablishedmethodformeasuringmolecularinteractions.TheirapplicationtothestudyofGPCRsisnonethelesslimitedtodetergent‐solubilizedreceptorsthatcanthenbereconstitutedintoalipidenvironment.UsingthechemokinereceptorCXCR4anditsspecificligandCXCL12,weoutlinehereahighlyreproduciblebiosensormethodbasedonreceptorpresentationonthesurfaceoflentiviralparticles;theapproachissimpleanddoesnotrequiretheuseofantibodiestoachievecorrectreceptororientationonthesensorchipsurface.WemeasuredthekineticparametersofCXCR4/CXCL12bindinginasinglestepandinrealtimeandevaluatedtheeffectofGAGpresentationofchemokinesonthisinteraction.Thedataindicatethatatlowconcentrations,solubleheparinmodulatesCXCR4/CXCL12interactionandathighconcentrations,abrogatesbinding.Theseobservationssuggestthatinadditiontotheirknownroleinmodulatinglocalchemokineavailability,GAGaffectthereceptor/ligandinteraction,althoughtheirinfluenceonaffinityparametersisverylimited.Themethodwillalsobeusefulforquantifyingthesebiomarkersinbiologicalfluidsandforthedevelopmentofhigh‐throughputscreeningfortheirantagonists...
