HIV‐infectedPMshowrestrictedreplicationascomparedwithMDM.WeaimedtodetermineatwhatpointintheviralreplicationcyclethisrestrictionoccursinPMascomparedwithMDM.WeperformedAlu‐LTRPCRforproviralDNAtodetectdifferencesinHIVintegration,real‐timeRT‐PCRtomeasureenvandgagmRNAlevels,andWesternblotanalysistodetectdifferencesinviralproteinexpression.PMandMDMwereinfectedwithHIV‐1BaL,andDNAwasextractedafter24handat6daysp.i.forreal‐timePCRstudies.At6and12daysp.i.,cellswerelysedforWesternblotanalyses.WefoundnodifferenceinviralintegrationbetweenPMandMDMbutsignificantlylowerlevelsofviralproteingp120inPMthaninMDM.Real‐timeRT‐PCRanalysesrevealed24‐foldlessenvmRNAandtenfoldlessgagmRNAinPM.TheseresultssuggestthatHIV‐1restrictioninPMoccursattheleveloftranscription.Thisstudyissignificant,asitadvancesourunderstandingofHIV‐1infectioninPManditscontributiontodecreasedinuteroverticaltransmission...
