AsexpressionofCxsincellsoftheimmunesystemincreasesuponcellularactivation,weinvestigatedwhetherCxsandespeciallyCxHcsplayamajorroleduringTcell‐mediatedresponses.Inparticular,westudiedtheexpressionofCx43HcfollowingCD4+Tcellstimulationusingflowcytometry,real‐timePCR,andWesternblotanalysis.WeshowedthatexpressionofCx43anditsphosphorylatedisoformsincreasedinresponsetotheengagementofCD3andCD28.Cx43HcswerefoundtobeinvolvedinsustainingproliferationofTcells,asassessedbycellcyclestaining,thymidineincorporationassays,andCFSEanalysisofcellsexposedtomimeticpeptideinhibitorsoftheplasmamembraneCxchannelsandantibodiesgeneratedtoanextracellularregionofCx.ThereductionofTcellproliferationmediatedbyCxchannelinhibitorssuppressedcysteineuptakebutnotcytokineproduction.Weconcludethatuponantigenrecognition,TcellsrequireCxHctosustaintheirclonalexpansion...
