ThefunctionofneutrophilsintheinflammatoryresponseismodulatedbycontactwithECMproteins.WehavenowinvestigatedtheeffectofcollagentypeIonsecretionofthecytokineIL‐8byhumanneutrophilsinvitro.CollagentypeIinhibitedthesecretionofIL‐8fromneutrophilsmaintainedunderbasalconditionsorstimulatedwithfMLF.Thiseffectwasaccompaniedbydown‐regulationofIL‐8mRNA,anditappearedtobespecifictocollagentypeIamongECMproteins,inthatitwasnotobservedwithfibronectinorlaminin.TheinhibitoryeffectofcollagentypeIonIL‐8secretionwasdependentoncollagenconcentrationandcelldensity.Itwasalsoabolishedinthepresenceofantibodiestointegrinα2β1butwasnotaffectedbyantibodiestointegrinα5β1orβ4.OurresultsthussuggestthatcollagentypeIinhibitsthesecretionofIL‐8byhumanneutrophilsinaselectivemannerandthatthiseffectismediatedbytheinteractionofcollagenwithintegrinα2β1...
