IthasbecomeapparentthatregulationofROSisimportantincellsignalingandhomeostasis.AccumulationofROStriggersoxidativestressinvariouscelltypesandcontributestothedevelopment,progression,andpersistenceofcancer.RecentresearchhasdemonstratedthatredoxdysregulationcausedbyROSpromotesproliferation,differentiation,genomic,andepigeneticalterations;immuneevasion;andsurvivalinleukemiccells.ROSactassignalingmoleculestoregulateredox‐sensitivetranscriptionalfactors,enzymes,oncogenes,andotherdownstreameffectors.Thus,athoroughunderstandingtheroleofROSaskeymediatorsinleukemogenesisislikelytoprovideopportunitiesforimprovedpharmacologicalintervention.Inthisreview,wesummarizetherecentfindingsthatsupportaroleforROSinthepathogenesisofAMLandoutlineinnovativeapproachesintheimplementationofredoxtherapiesformyeloidmalignancies...
