【 摘 要 】
IncreasinginformationhasshownthatmanynewlyemergingstrainsofMycobacteriumtuberculosis,includingthehighlyprevalentandtroublesomeBeijingfamilyofstrains,canpotentlyinducetheemergenceofFoxp3+CD4Tregs.Althoughthesignificanceofthisisstillnotfullyunderstood,wehavepreviouslyprovidedevidencethattheemergenceofthispopulationcansignificantlyablatetheprotectiveeffectofBCGvaccination,causingprogressivefataldiseaseinthemousemodel.However,whetherthepurposeofthisresponseistocontrolinflammationortodirectlydampentheacquiredimmuneresponseisstillunclear.Inthepresentstudy,wehaveshown,usingbothcelldepletionandadoptivetransferstrategies,thatTregscanhaveeitherproperties.Celldepletionresultedinarapid,buttransient,decreaseinthelungbacterialload,suggestingreleaseortemporaryre‐expansionofeffectorimmunity.TransferofTregsintoRag2−/−ormarkedcongenicmiceworsenedthediseasecourseanddepressedcellularinfluxofeffectorTcellsintothelungs.Tregsfrominfecteddonorsseemedtopreferentiallydepresstheinflammatoryresponseandgranulocyticinflux.Incontrast,thosefromBCG‐vaccinatedandthenchallengeddonorsseemedmorefocusedondepressionofacquiredimmunity.ThesequalitativedifferencesmightberelatedtoincreasingknowledgereflectingtheplasticityoftheTregresponse...
