HIV‐1‐infectedsubjects,despitecontrolofviralreplicationwithART,haveanalteredimmunecytokine/chemokinemilieu.Changesinsystemiccytokinesandchemokinescanalterimmuneresponses.IP‐10,inparticular,hasbeenassociatedwithpathogenesisinanumberofconditions,andwefoundthatIP‐10isincreasedinseruminsubjectswhoareHIV‐1infectedandonstableARTcomparedwithHIV‐1‐uninfectedindividuals.Inaseriesofinvitrostudies,wefoundthatPBMCsexposedtoIP‐10showedasignificantdecreaseinthenumberofcellscapableofsecretingIFN‐γ,aswellasothercytokines,whenstimulatedwithrecallantigens.Furthermore,treatmentwithIP‐10ledtodecreasedantigen‐specificcalciumsignalingandMAPK38phosphorylation.Importantly,thecytokines,aswellasproliferativeresponses,couldbeenhancedwithanIP‐10Nab.OurfindingssuggestthatIP‐10‐modulatingdrugsmaypotentiallyenhanceTcellresponsestovaccinationandHIV‐1inHIV+subjectsonART...【 授权许可】
