CD200isatransmembraneproteinbroadlyexpressedonavarietyofcelltypes,whichdeliversimmunoregulatorysignalsthroughbindingtoreceptors(CD200Rs)expressedonmonocytes/myeloidcellsandTlymphocytes.SignalsdeliveredthroughtheCD200:CD200Raxishavebeenshowntoplayanimportantroleintheregulationofanti‐tumorimmunity,andoverexpressionofCD200hasbeenreportedinanumberofmalignancies,includingCLL,aswellasoncancerstemcells.WeinvestigatedtheeffectofCD200blockadeinvitroonagenerationofCTLresponsesagainstapoorlyimmunogenicCD200+lymphomacelllineandfreshcellsobtainedfromCLLpatientsusinganti‐CD200mAbandCD200‐specificsiRNAs.SuppressionoffunctionalexpressionofCD200augmentedkillingoftheCD200+cells,aswellasproductionoftheinflammatorycytokinesIFN‐γandTNF‐αbyeffectorPBMCs.KillingwasmediatedbyCD8+cytotoxicTcells,andCD4+TcellsplayanimportantroleinCD200‐mediatedsuppressionofCTLresponses.OurdatasuggestthatCD200blockademayrepresentanovelapproachtoclinicaltreatmentofCLL...
