Acutesystemicinflammationfrominfectiousandnoninfectiousetiologieshasstereotypicfeaturesthatprogressthroughaninitiation(proinflammatory)phase,anadaptive(anti‐inflammatory)phase,andaresolution(restorationofhomeostasis)phase.Thesephase‐shiftsareaccompaniedbyprofoundandpredictablechangesingeneexpressionandmetabolism.Here,wereviewtheemergingconceptthatthetemporalphasesofacutesystemicinflammationarecontrolledbyanintegratedbioenergyandepigeneticbridgethatguidesthetimingoftranscriptionalandpost‐transcriptionalprocessesofspecificgenesets.Thisunifyingconnectiondepends,atleastinpart,onredoxsensorNAD+‐dependentdeacetylase,Sirt1,andaNF‐κB‐dependentp65andRelBfeed‐forwardandgene‐specificpathwaythatgeneratessilentfacultativeheterochromatinandactiveeuchromatin.Anadditionallevelofregulationforgene‐specificreprogrammingisgeneratedbydifferentialexpressionofmiRNAthatdirectlyandindirectlydisruptstranslationofinflammatorygenes.Thesemolecularreprogrammingcircuitsgenerateadynamicchromatinlandscapethattemporallydefinesthecourseofacuteinflammation...
