ThisstudyinvestigatedinvivotheinfluenceofageandvitaminDstatusoninnateimmunefunctioninHC.Serum25OHDwasmeasuredin71HC.TLRexpressiononvarioussubpopulationsofPBMCs,aswellasTLRfunctionbystimulatingPBMCswithspecificligands,wasassessedbyflowcytometry.CirculatingcathelicidinlevelsweredeterminedbyELISA.Serum25OHDlevelsdecreasedwithage,andtherewasasignificantinversecorrelationbetween25OHDlevelsandage.Therewasanegativecorrelationbetweenserum25OHDlevelsandMFIexpressionofTLR7onBcells,Tcells,andmonocytes.TLR7function,addressedbyinvitrostimulationwithaspecificagonist,wassignificantlycorrelatedwithserum25OHDlevels,andthiswasespeciallyaresultoftheresultsinHColderthan60years.MFIexpressionofTLR5onTcellsandTLR2onmonocyteswasalsonegativelycorrelatedwithserum25OHDlevels.TLR1(monocytes)andTLR2(monocytes)expressionwaspositivelycorrelatedwithage.Furthermore,TLR4andTLR8functionwasnegativelycorrelatedwithage.Circulatingcathelicidinlevelsdecreasedwithageandwerepositivelycorrelatedwith25OHDlevels.AgingisaccompaniedbychangesinexpressionandfunctionofseveralTLRs.Serum25OHDlevelsdecreasewithageandarealsoassociatedwithachangeinexpressionanddefectivefunctionofcertainTLRs,especiallythoseinvolvedinviralresponse...
