TLRsplayamajorroleinmicrobe‐hostinteractionsandinnateimmunity.Ofthe10functionalTLRsdescribedinhumans,TLR2isuniqueinitsrequirementtoformheterodimerswithTLR1orTLR6fortheinitiationofsignalingandcellularactivation.TheligandspecificityofTLR2heterodimershasbeenstudiedextensively,usingspecificbacterialandsyntheticlipoproteinstogaininsightintothestructure‐functionrelationship,theminimalactivemotifs,andthecriticaldependenceonTLR1orTLR6foractivation.Differentfromthatforspecificwell‐definedTLR2agonists,recognitionofmorecomplexligandslikeintactmicrobesormoleculesfromendogenousoriginrequiresTLR2tointeractwithadditionalcoreceptors.Abreadthofdatahasbeenpublishedonligand‐inducedinteractionsofTLR2withadditionalpatternrecognitionreceptorssuchasCD14,scavengerreceptors,integrins,andarangeofotherreceptors,allofthemimportantfactorsinTLR2function.ThisreviewsummarizestherolesofTLR2invivoandinspecificimmunecelltypesandintegratesthisinformationwithadetailedreviewofourcurrentunderstandingoftherolesofspecificcoreceptorsandligandsinregulatingTLR2functions.Understandinghowtheseprocessesaffectintracellularsignalinganddrivefunctionalimmuneresponseswillleadtoabetterunderstandingofhost‐microbeinteractionsandwillaidinthedesignofnewagentstotargetTLR2functioninhealthanddisease...
