HMGB1isanarchitecturalchromatin‐bindingproteinthatcanbereleasedactivelybyactivatedcellsorpassivelybydyingcellsandcanserveasaDAMPmoleculetodrivethepathogenesisofinflammatoryandangiogenicdiseases.ThroughTLR4andRAGEsignalingpathways,HMGB1couldregulatevasculargrowthinvivoandinvitrothroughdiversemechanisms,includinginductionofproangiogeniccytokinereleaseandactivationofECs,macrophages,EPCs,andmesoangioblasts,allofwhichcouldcontributetovesselformation.Accordingly,HMGB1playsasignificantroleinmanyangiogenesis‐relatedconditions,suchastumors,PDR,wound‐healing,andischemia‐inducedangiogenesis.Inthisreview,wefocusontheregulatoryroleofHMGB1inangiogenesisandrecentprogressintherapeuticstrategiestargetingHMGB1...
