【 摘 要 】
TheefficacyofdonorHSCTispartlyreducedasaresultofslowpost‐transplantationimmunerecovery.Inparticular,Tcellregenerationisgenerallydelayed,resultinginhighinfection‐relatedmortalityinthefirstyearspost‐transplantation.Adoptivetransferofinvitro‐generatedhumanTcellprogenitorsseemsapromisingapproachtoaccelerateTcellrecoveryinimmunocompromisedpatients.AAmayenhanceTcellproliferationanddifferentiationinacontrolled,feeder‐freeenvironmentcontainingNotchligandsanddefinedgrowthfactors.OurexperimentsshowapivotalroleforAAduringhumaninvitroTcelldevelopment.TheblockingofNOSdiminishedthiseffect,indicatingaroleforthecitrulline/NOcycle.AApromotesthetransitionofproT1toproT2cellsandofpreTtoDPTcells.Furthermore,theadditionofAAtofeedercoculturesresultedindevelopmentofDPandSPTcells,whereaswithoutAA,apreTcell‐stagearrestoccurred.WeconcludethatneitherDLL4‐expressingfeedercellsnorfeedercellconditionedmediaarerequiredforgeneratingDPTcellsfromCBandG‐CSF‐mobilizedHSCsandthatgenerationandproliferationofproTandDPTcellsaregreatlyimprovedbyAA.ThistechnologycouldpotentiallybeusedtogenerateTcellprogenitorsforadoptivetherapy...
