【 摘 要 】

pCHisanimportantriskfactorforbraininjuryandlong‐termmorbidityinchildren,occurringduringthedevelopmentalstagesofneurogenesis,neuronalmigration,andmyelination.WeshowthatarodentmodelofpCHresultsinanearlydecreaseinmaturemyelin.AlthoughpCHdoesincreaseprogenitoroligodendrocytesinthedevelopingbrain,BrdUlabelingrevealedalossindividingprogenitoroligodendrocytes,indicatingadefectinmaturecellreplacementandmyelinogenesis.Micecontinuedtoexhibitedhypomyelination,concomitantwithlong‐termimpairmentofmotorfunction,weeksaftercessationofpCH.Theimplicationofanovelneuroimmunologicinterplay,pCHalsoinducedasignificantegressofinfiltratingCD4Tcellsintothedevelopingbrain.ThispCH‐mediatedneuroinflammationincludedoligodendrocyte‐directedautoimmunity,withanincreaseinperipheralmyelin‐specificCD4Tcells.Thus,boththelossofavailable,mature,myelin‐producingglialcellsandanactiveincreaseinautoreactive,myelin‐specificCD4TcellinfiltrationintopCHbrainsmaycontributetoearlypCH‐inducedhypomyelinationinthedevelopingCNS.Theelucidationofpotentialmechanismsofhypoxia‐drivenautoimmunitywillexpandourunderstandingoftheneuroimmuneaxisduringperinatalCNSdiseasestatesthatmaycontributetolong‐termfunctionaldisability...
