HumanCD4+CD25hiFOXP3+regulatoryTcellsmaintainimmunologictoleranceandpreventautoimmuneandinflammatoryimmuneresponses.RegulatoryTcellsundergoasimilaractivationcycleasconventionalCD4+Tcellsuponantigenstimulation.Here,wedemonstratethatTcellreceptorsandcostimulationarerequiredtoactivatetheregulatoryTcellsuppressivefunction.RegulatoryTcellssuppressedtheTcellreceptorsignalingineffectorTcellsinatime‐dependentmannerthatcorrespondedwithinhibitionofcytokineproductionandproliferation.ModulationoftheactivationlevelandtherebythesuppressivecapacityofregulatoryTcellsimposeddistinctTcellreceptorsignalingsignaturesandhyporesponsivenessinsuppressedandproliferatingeffectorTcellsandestablishedathresholdforeffectorTcellproliferation.TheimmunesuppressionofeffectorTcellswascompletelyreversibleuponremovalofregulatoryTcells.However,thestrengthofpriorimmunesuppressionbyregulatoryTcellsandcorrespondingTcellreceptorsignalingineffectorTcellsdeterminedthesusceptibilitytosuppressionuponlaterreexposuretoregulatoryTcells.ThesefindingsdemonstratehowthestrengthoftheregulatoryTcellsuppressivefunctiondeterminesintracellularsignaling,immuneresponsiveness,andthelatersusceptibilityofeffectorTcellstoimmunesuppressionandcontributetounveilingthecomplexinteractionsbetweenregulatoryTcellsandeffectorTcells...
