Whereasadenoviralvectorsareknowntoactivatethecomplementcascade,leadingtofixationofC3proteinstotheviralcapsid,theconsequencesofthisactivationforviralclearancefromthecirculationarenotknown.LiverKCs,themacrophagepopulationresponsibleforearlyuptakeandeliminationofmanyblood‐bornepathogens,expressCRIg,acomplementreceptorforC3proteins.Here,wefindthatCRIgisimportantfortheearlyeliminationofC3‐coatedadenoviralvectorsfromthesinusoidalbloodstreambyKCs.Wefurtherdemonstratethatbyactingasacriticalreceptorforadenovirusphagocytosis,CRIgplaysanimportantroleinregulatingvirus‐inducedKCdeathanddepletionofthesecellsfromtheliversinusoidallumen.OurstudythusidentifiesacriticalpathwayregulatingKCfunctionandsurvivalinresponsetosystemicviralinfection...
