Theplasmamembraneofimmunecellsisahighlyorganizedcellstructurethatiskeytotheinitiationandregulationofinnateandadaptiveimmuneresponses.Itiswell‐establishedthatimmunoreceptorsembeddedintheplasmamembranehaveanonrandomspatialdistributionthatisimportantforcouplingtocomponentsofintracellularsignalingcascades.Inthelasttwodecades,specializedmembranemicrodomains,includinglipidraftsandTEMs,havebeenidentified.Thesedomainsarepreformedstructures(“physicalentities”)thatcompartmentalizeproteins,lipids,andsignalingmoleculesintomultimolecularassemblies.InAPCs,differentmicrodomainscontainingimmunoreceptors(MHCproteins,PRRs,integrins,amongothers)havebeenreportedthatareimperativeforefficientpathogenrecognition,theformationoftheimmunologicalsynapse,andsubsequentTcellactivation.Inaddition,recentworkhasdemonstratedthattetraspaninmicrodomainsandlipidraftsareinvolvedinBCRsignalingandBcellactivation.Researchintothemolecularmechanismsunderlyingmembranedomainformationisfundamentaltoacomprehensiveunderstandingofmembrane‐proximalsignalingandAPCfunction.Thisreviewwillalsodiscusstheadvancesinthemicroscopyfieldforthevisualizationoftheplasmamembrane,aswellastherecentprogressintargetingmicrodomainsasnovel,therapeuticapproachforinfectiousandmalignantdiseases...
