【 摘 要 】
Epigeneticscontributestothedevelopmentofvarietyofdiseasesbymodulationofgeneexpression.WeevaluatedtheeffectofHIV‐inducedVDRmethylationonlossofTCs.HIV/TCdisplayedenhancedVDR‐CpGmethylationandincreasedexpressionofDnmt3bbutattenuatedexpressionofVDR.Ademethylatingagent,AZA,inhibitedthiseffectofHIV.HIV/TCalsodisplayedtheactivationoftheRAS,whichwasreversedbyEB(aVDA).Further,HIV/TCsdisplayedenhancedgenerationofROSandinductionofDSBsbutattenuatedDNArepairresponse.However,inthepresenceofAZA,EB,LOS(aRASblocker),Cat,andtempol(freeradicalscavengers),HIV‐inducedTCROSgenerationandinductionofDSBswereattenuatedbutassociatedwithenhancedDNArepair.Additionally,AZA,EB,andLOSprovidedprotectionagainstHIV‐inducedTCapoptosis.ThesefindingssuggestedthatHIV‐inducedTCapoptosiswasmediatedthroughROSgenerationinresponsetoHIV‐inducedVDRmethylationandassociatedactivationoftheRAS...
