TheroleofJNKinneutrophilchemotaxisandkillingofmicrobialpathogensremainsunclear.Usingarecentlydescribedcell‐permeablepeptideinhibitoroftheJNKpathway,basedontheJBDofJIP‐1,coupledtotheproteintransductiondomainofHIV‐TAT(TAT‐JIP),inassociationwithcontrolpeptides,wedemonstratethattheJNKpathwayplaysamajorroleinregulatinghumanneutrophilchemotaxisandkillingofmicrobialpathogens.Serum‐opsonizedStaphylococcusaureuselicitedJNKactivationandc‐junphosphorylation.TheactivationoftheJNKpathwayandbactericidalactivitywereinhibitedbytheTAT‐JIPpeptide.ThestimulationofoxygenradicalgenerationbyS.aureuswasdependentontheJNKsignalingpathway,aswasthephagocytosisofserum‐opsonizedbacteria.ChemotaxistoactivatedserumcomplementbutnotrandommigrationwasinhibitedbytheTAT‐JIPpeptide.ThefindingsdemonstrateamajorrolefortheJNKsignalingpathwayinneutrophil‐mediateddefenseagainstmicrobialpathogens...
