Cytoskeletalrearrangementsoftenoccurastheresultoftransductionofsignalsfromtheextracellularenvironment.Efficientawakeningofthispowerfulmachineryrequiresmultipleactivationanddeactivationsteps,whichusuallyinvolvephosphorylationordephosphorylationofdifferentsignalingunitsbykinasesandphosphatases,respectively.Inthisreview,wediscussthesignalingcharacteristicsofoneofthenPKCisoforms,PKCθ,focusingonPKCθ‐mediatedsignaltransductiontocytoskeletalelements,whichresultsincellularrearrangementscriticalforcelltype‐specificresponsestostimuli.PKCθisthemajorPKCisoformpresentinhematopoieticandskeletalmusclecells.PKCθplaysrolesinTcellsignalingthroughtheIS,survivalresponsesinadultTcells,andTcellFasL‐mediatedapoptosis,allofwhichinvolvecytoskeletalrearrangementsandrelocationofthisenzyme.PKCθhasbeenlinkedtotheregulationofcellmigration,lymphoidcellmotility,andinsulinsignalingandresistanceinskeletalmusclecells.AdditionalrolesweresuggestedforPKCθinmitosisandcell‐cycleregulation.Comprehensiveunderstandingofcytoskeletalregulationandthecellular“modusoperandi”ofPKCθholdspromiseforimprovingcurrenttherapeuticapplicationsaimedatautoimmunediseases...
