DCsplayacentralroleinthedevelopmentofinnateandadaptiveimmunitybutalsointheinductionandmaintenanceofimmunetolerance.IdentificationoffactorsthatgovernDCactivation,theirmaturationstate,andtheircapacitytoinduceproinflammatoryortolerogeneicresponsesthereforerepresentsacrucialaimofresearch.Wepreviouslyidentifiedanewmolecule,Tmem176B(whichwenamedTORIDinitially),ashighlyexpressedinamodelofallografttoleranceintherat.WeshowedthatitsoverexpressioninratDCsblockedtheirmaturation,suggestingaroleforthismoleculeinthematurationprocess.TocharacterizethefunctionofTmem176Bfurther,weusedasplit‐ubiquitinyeast,two‐hybridsystemtoidentifyinteractingpartnersandfoundthatTmem176BassociatedwithitselfbutalsowithTmem176A,amembraneproteinsimilartoTmem176B.Interestingly,thesetwomoleculesshowedsimilarmRNAexpressionpatternsamongvariousmurinetissuesandimmunecellsandwerebothdown‐regulatedfollowingDCmaturation.Inaddition,weshowedthatinusingRNAi,thesemoleculesarebothinvolvedinthemaintenanceoftheimmaturestateoftheDCs.Takentogether,thesedatasuggestthatTmem176BandTmem176AassociatetoformmultimersandrestrainDCmaturation.Therefore,thesetwomoleculesmayrepresentvalidtargetstoregulateDCfunction...
