TheHVEM,orTNFRSF14,isamembrane‐boundreceptorknowntoactivatetheNF‐κBpathway,leadingtotheinductionofproinflammatoryandcellsurvival‐promotinggenes.HVEMbindsseveralligandsthatarecapableofmediatingcostimulatorypathways,predominantlythroughitsinteractionwithLIGHT(TNFSF14).However,itcanalsomediatecoinhibitoryeffects,predominantlybyinteractingwithIGSFmembers,BTLAorCD160.Therefore,itcanfunctionlikea“molecularswitch”forvariousactivatingorinhibitoryfunctions.Furthermore,recentstudiessuggesttheexistenceofbidirectionalsignalingwithHVEMactingasaligandforsignalingthroughBTLA,whichmayactasaligandinothercontexts.Bidirectionalsignaling,togetherwithnewinformationindicatingsignalingincisbycellsthatcoexpressHVEManditsligands,makessignalingwithinaHVEM‐mediatednetworkcomplicated,althoughpotentiallyrichinbiology.AccumulatinginvivoevidencehasshownthatHVEM‐mediated,coinhibitorysignalingmaybedominantoverHVEM‐mediatedcostimulatorysignaling.InseveraldiseasemodelstheabsenceofHVEM‐BTLAsignalingpredominantlyresultedinseveremucosalinflammationinthegutandlung,autoimmune‐likedisease,andimpairedimmunityduringbacterialinfection.Here,wewillsummarizethecurrentviewabouthowHVEM‐BTLAsignalingisinvolvedintheregulationofmucosalinflammation,autoimmunity,andinfectionimmunity...
