ThePRRTLR7playsakeyroleintheactivationofautoantigen‐reactiveBcells.ThisresponseisincreasedmarkedlybyIFN‐α,producedbyaccessorycells,asaresultoftheup‐regulationofTLR7.WereporthereinanalternativepathwaybywhichTLR7expressioncanbeaugmented.ThisfindingwasderivedfromcontinuationofongoingstudiestouncoverinteractionsbetweenNKandBcells.Here,wehavecomparedgeneexpressionprofilesbymicroarrayanalysisofBcellsbeforeandaftertheirinteractionwithpurifiedNKcells.ThemostoutstandingalterationofgenestranscribedinBcellsisasignificantincreaseintheexpressionofmanymembersoftheISGfamily,amongwhichisTLR7.FurtheranalysisrevealedthattheenhancementofTLR7onBcellsisnotmediatedviatypeIortypeIIIFNbutbyanothercytokine,IL‐28,atypeIIIIFN,whichactsinconcertwithcontact‐mediatedinteractionswithNKcells.ThisincreasedexpressionallowsBcellstorespondmorereadilyuponstimulationbyitsligandandmayincreaseinvivoresponsestootherTLR7ligands,suchasautoantigens,priortoorjointlywithstimulationbyothercytokines...
