Originallydescribedin2002asaTcell‐costimulatorycytokine,thetumornecrosisfactorfamilymemberTNF‐likefactor1A(TL1A),encodedbytheTNFSF15gene,hassincebeenfoundtoaffectmultiplecelllineagesthroughitsreceptor,deathreceptor3(DR3,encodedbyTNFRSF25)withdistinctcell‐typeeffects.GeneticdeficiencyorblockadeofTL1A‐DR3hasdefinedanumberofdiseasestatesthatdependonthiscytokine‐receptorpair,whereasexcessTL1Aleadstoallergicgastrointestinalinflammationthroughstimulationofgroup2innatelymphoidcells.NoncodingvariantsintheTL1Alocusareassociatedwithsusceptibilitytoinflammatoryboweldiseaseandleprosy,predictingthatthelevelofTL1Aexpressionmayinfluencehostdefenseandthedevelopmentofautoimmuneandinflammatorydiseases...
