TheCD70/CD27axishasgainedincreasinginterestamongtheimmunologists,becauseofitscapacitytoregulateimmunityversustolerance.RecentstudiesclearlyshowthatexpressionofCD70maypreventtoleranceinducedbyantigenpresentationinthesteady‐state,i.e.,bynonactivatedDCs.Inaddition,CD27signalingappearscriticalforTcellexpansionandsurvivalandtherefore,inductionoflong‐termmemory.Itcontributestogerminalcenterformation,Bcellactivation,andproductionofneutralizingantibodiesbutcanalsobesubvertedbyviruses,inparticular,duringchronicinfections.ThepotentialroleoftheCD27/CD70pathwayinthecourseofinflammatorydiseases,asinEAE,arthritis,andinflammatoryboweldiseasemodels,suggeststhatCD70maybeatargetforimmuneintervention.Conversely,thepotencyofcostimulationthroughCD27suggeststhattheCD27/CD70axiscouldbeexploitedforthedesignofanti‐cancervaccines...
