Inthephagocytosisfield,ROSproductionbythephagocyteNOXhasbeenassociatedwithpathogenkillingforthelast50years.SincethediscoveryofnonphagocyteNOX,numerousotherrolesforROSproductionhavebeenidentified.OxidativestressandROS‐mediatedsignalinghavereceivedmuchattentioninrecentyears.MuchlowerconcentrationsofROSmayberequiredforsignalingcomparedwithmicrobialkilling.Basedonthediscoveriesinnonphagocyticcells,itbecamelogicaltolookforROSfunctionsdistinctfrompathogenkilling,eveninphagocytes.ROSarenowlinkedtovariousformsofcelldeath,tochemotaxis,andtonumerousmodificationsofcellularprocesses,includingtheNOXitself.ROSfunctionsareclearlyconcentration‐dependentoverawiderangeofconcentrations.Howmuchisrequiredforwhichfunction?Whichspeciesarerequiredforhowmuchtime?IsROSsignalingonlyasideeffectofbactericidalROSproduction?OnemajorobstacletoanswerthesequestionsisthedifficultyofreliablequantitativeROSdetection.Signaltransductionoftentakesplaceonasubcellularscaleoverperiodsofsecondsorminutes,sothedetectionmethodsneedtoprovideappropriatetimeandspaceresolution.WepresentexamplesoflocalROSproduction,decreaseddegradation,signalingevents,andpotentiallyROS‐sensitivefunctions.WeattempttoillustratethecurrentlimitationsforquantitativespatiotemporalROSdetectionandpointoutdirectionsforongoingdevelopment.ProbesforlocalizedROSdetectionandforcombineddetectionofROS,togetherwithproteinlocalizationorothercellularparameters,areconstantlyimproved...
