AhallmarkofLListheaccumulationofVirchow'sfoamymacrophages.However,theoriginandnatureoftheselipids,aswellastheirfunctionandcontributiontoleprosydisease,remainunclear.WehereinshowthatmacrophagespresentinLLdermallesionsarehighlypositiveforADRP,suggestingthattheirfoamyaspectisatleastinpartderivedfromLD(alsoknownaslipidbodies)accumulationinducedduringMLinfection.Indeed,thecapacityofMLtoinduceLDformationwasconfirmedinvivoviaanexperimentalmodelofmousepleurisyandininvitrostudieswithhumanperipheralmonocytesandmurineperitonealmacrophages.Furthermore,infectedcellswereshowntopropagateLDinductiontouninfected,neighboringcellsbygeneratingaparacrinesignal,forwhichTLR2andTLR6weredemonstratedtobeessential.However,TLR2andTLR6deletionsaffectedLDformationinbacterium‐bearingcellsonlypartially,suggestingtheinvolvementofalternativereceptorsoftheinnateimmuneresponsebesidesTLR2/6forMLrecognitionbymacrophages.Finally,adirectcorrelationbetweenLDformationandPGE2productionwasobserved,indicatingthatML‐inducedLDsconstituteintracellularsitesforeicosanoidsynthesisandthatfoamycellsmaybecriticalregulatorsinsubvertingtheimmuneresponseinleprosy...
