ImmuneresponsesagainstPlasmodiumparasites,thecausativeorganismsofmalaria,aretraditionallydichotomizedintopre‐erythrocyticandblood‐stagecomponents.Whereasthecentralroleofcellularresponsesinpre‐erythrocyticimmunityiswellestablished,protectionagainstblood‐stageparasiteshasgenerallybeenascribedtohumoralresponses.Anumberofrecentstudies,however,havehighlightedtheexistenceofcellularimmunityagainstblood‐stageparasites,inparticular,theprominenceofIFN‐γproduction.Here,wehaveundertakentochartthecontributionofthisprototypicalcellularcytokinetoimmunityagainstpre‐erythrocyticandblood‐stageparasites.WesummarizethevariousantiparasiticeffectorfunctionsthatIFN‐γservestoinduce,reviewanarrayofdataaboutitsprotectiveeffects,andscrutinizeevidenceforanydeleterious,immunopathologicaloutcomeinmalariapatients.WediscusstheactivationandcontributionofdifferentcellularsourcesofIFN‐γproductionduringmalariainfectionanditsregulationinrelationtoexposure.WeconcludethatIFN‐γformsacentralmediatorofprotectiveimmuneresponsesagainstpre‐erythrocyticandblood‐stagemalariaparasitesandidentifyanumberofimplicationsforrationalmalariavaccinedevelopment...
