Forleukocytestopenetratethevesselwall,theyneedtointeractsequentiallywiththeendothelialliningandtheperivascularBM.Thematrixproteinlaminin‐411isamajorconstituentofthevascularBM.Thelamininα4chainisacomponentoflaminin‐411andhasstructuralandsignalingfunctions.Here,weaddressedtheroleofBMlamininα4inleukocyterecruitmenttoinflammatoryloci.WeusedseveralrecruitmentmodelsinLam4−/−andWTmicetodeterminewhetherlackoflaminin‐411intheperivascularBMinfluencesextravasationofinflammatorycells.Recruitmentofallmajorleukocytesubsets(neutrophils,monocytes,andlymphocytes)wasreducedinLam4−/−micecomparedwithWT.Withtheuseofintravitalmicroscopy,weconcludedthatthisdecreasewasaresultofimpaireddiapedesisthroughthevesselwall,asneitherleukocyteadhesiontotheendothelialliningnormigrationinextravasculartissuewashamperedinLam4−/−mice.Collectively,ourdatasuggestareducedabilityofimmunecellstopenetratethevesselwallinmicedeficientinlamininα4...
