【 摘 要 】

Neutrophilβ1integrinexpressionandcontributiontocelladhesionwererevisitedinthisstudy.α9β1andα5β1appearedhereasthemainβ1integrinsexpressedonthemembraneofrestingplatelet‐depletedneutrophils—α6β1representing<15%andα2β1undetectable.Neutrophilactivationslightlyenhancedα5expression,didnotchangeα6,butresultedinatwo‐tothreefoldincreaseofα9β1,whichthenbecamethemajorβ1integrinoftheneutrophilmembrane.α9β1wastheonlyβ1integrintobeup‐regulatedaftertransendothelialmigrationacrossTNF‐α‐activatedHUVECs.Asα9β1bindsVCAM‐1,weanalyzeditsparticipationtoneutrophiladhesiontoTNF‐α‐activatedendothelialcells.Blockinganti‐α9mAbhadlittleeffectonneutrophilstaticadhesion,contrastingwiththestronginhibitionbyanti‐β2mAb.Underflowconditions,theanti‐α9mAbhadnoeffectbyitselfonneutrophiladhesiontoactivatedHUVECsbutenhancedtheblockingeffectofanti‐β2antibodiessignificantlyandfurtherenhancedthevelocityofβ2–blockedrollingneutrophils.Inconclusion,wedescribehereforthefirsttimeanearlyexclusiveup‐regulationofα9β1expressionamongallβ1integrinsduringneutrophilactivationandtransendothelialmigrationandapossiblyimportantsynergybetweenα9β1andβ2integrinsinstabilizingneutrophiladhesiontoendotheliumunderflowconditions...
