Theserineproteaseplasmingeneratedfromitszymogenplasminogenisbestknownforitsfunctionasakeyenzymeofthefibrinolyticcascade.However,beyondfibrinolysis,plasminhasanumberofcrucialfunctionsinavarietyofprocesses,includinginflammation.Variouscellscanbindplasminogenandplasminviaplasminogen‐bindingsitesexposingaC‐terminallysine.Plasmin,generatedasaresultofplasminogenactivationatthecellsurface,isprotectedfromitsphysiologicalinhibitors.Apartfromitsabilitytofacilitatecellmigrationintissues,plasminiscapableoftriggeringsignaling,whichdependsoncellularbindingviaitslysine‐bindingsitesanditsproteolyticactivity.Plasmin‐inducedsignalingaffectsvariousfunctionsofmonocytes,macrophages,DCs,andothers,withthelistofaffectedcellsstillgrowing.Invitroandinvivostudieshavedemonstratedtheabilityofplasmintostimulatetheproductionofcytokines,ROS,andothermediators,therebycontributingtoinflammation.Plasmin‐inducedchemotaxisofmonocytesandDCsindicatesthatitisalsoapotentchemoattractantforimmunecells.Therefore,excessiveactivationofplasmininchronicinflammatoryorautoimmunediseasesmightexacerbatetheactivationofinflammatorycellsandthepathogenesisofthedisease.Thisreviewfocusesontheavailableevidenceforphysiologicalandpathophysiologicalrolestheserineproteaseplasminininflammatoryprocesses...
