DuringCP,thegingivalenvironmentisprimedtorecruitandactivatehominglymphocytes.However,detailedphenotypicandfunctionalcharacterizationofgingivaltissueresidentlymphocyteshasbeenchallengingasaresultoflimitationsassociatedwithavailableisolationmethodsandlimitedavailabilityofhumansamples.Thisstudyaimedtodevelopanovelexplantculturetechniqueforeffectivelyisolatinghumangingivallymphocytes.ThistechniquetakesadvantageofthenaturaltendencyofMNCstomigratetowardachemokinegradientgeneratedbythegingivalfibroblasts.TheexplantsystemallowedisolationofMNCswith∼95×higheryieldrelativetoconventionalapproaches.TheMNCyieldcorrelatesdirectlywithwetweightsofthetissues,andmaximalMNCsareisolatedduringthe2nddayoftheculture.TheexplantculturemethodandconventionalapproachproducesimilarMNCsubpopulationssuchasTh,Tc,andBcells.ChemokinesassociatedwithMNCmigrationbutnotcytokinesassociatedwithMNCproliferationanddifferentiationwereproducedintheexplantculture.Moreover,MNCmigrationinresponsetothesecretedchemokineswasinhibitedbyPTX.Tcellsdidnotundergoproliferationduringtheculture.However,theisolatedTcellsrespondedtomitogenicstimulationexvivo.Astatistically,significantlydecreasedThcellwithreducedCD25expressionalongwithincreasedNKandNKTcellsinCPareshown.However,thenumberofnaïveNKcellswasdecreasedsignificantlyinMNCs,suggestingactivationofNKcellsinCP...
