Neutrophilsareessentialeffectorcellsinhostdefenseagainstinvadingpathogens.Regulationofadhesion,migration,andchemotacticprocessesisimportantinthehomingandactivationofthesecells.Werecentlydescribedthreedistinctsubsetsofcirculatinghumanneutrophilsinperipheralbloodduringacutesystemicinflammation.Onesubset,CD16bright/CD62Ldim,hasimmunesuppressivecharacteristicsbecauseitcaninhibitT‐cellproliferation.TheothertwosubsetsconsistofbandedCD16dim/CD62Lbrightandphenotypicallymature(normal)CD16bright/CD62Lbrightneutrophils.Thecurrentstudywasdesignedtodeterminetheadhesioncharacteristicsofthesedifferentneutrophilsubsets.AnalysisofadhesiontoactivatedendotheliumunderflowconditionsrevealedthatCD16bright/CD62LdimneutrophilsadheredlesscomparedwithCD16bright/CD62LbrightandCD16dim/CD62Lbrightneutrophils.ThisdecreaseinbindingcapacitycouldbemimickedintheotherneutrophilsubsetsbyblockingL‐selectin.ChemotaxisofCD16bright/CD62Ldimneutrophilstotheend‐targetchemoattractantN‐formylmethionine‐leucine‐phenylalaninewaslowercomparedwiththatfortheCD16dim/CD62Lbrightneutrophilsubset,whereaschemotaxistocell‐derivedchemoattractantCXCL8wascomparable.Ourdataindicatethatcaptureonendotheliumunderflowconditions,akeymechanismnecessaryforextravasation,ofCD16bright/CD62Ldimneutrophilstoinflammatorysitesisattenuated,whichmayfacilitatemigrationofthesecellstoothertissuelocalizations.Modulationofthisprocessisapotentialtargettomanipulateinflammationbecausepotentiationofthisimmunesuppressionmightaidinanti‐inflammatorytherapy...
