Decidualization,aprogesterone‐dependentprocessthataltersendometrialstromalcellsatimplantationsitesinhumansandrodents,isaccompaniedbyahighlyregulated,NKcell‐dominatedleukocyteinfluxintodecidualbasalis(DB).WhetheruNKcellsdifferentiatefromuterineprogenitorcellsisunknown,asarethemechanismsrestrictingleukocytestoDB.WeaskedifcellsexpressingtheearlyNKlineagemarkerCD127(IL‐7Rα)occurredinmousedecidua.CD127wasabsentfromgd6.5deciduallymphoidcellsbutbecameexpressedbyamatureuNKcellsubsetingd10.5DB.DBandtransientmyometrialstructures(MLAp)thatringmaternalbloodvesselssupplyingplacentaeexpressedIL‐7andTSLP,theCD127ligands,butwithdifferingtemporalandspatialpatterns.UNKcellsexpressedTSLPR,andstudyofgd10.5implantationsitesfrommicedeletedforIL‐7,CD127,orTSLPRsuggestedthatIL‐7anditsreceptorhavephysiologicalrolesinlimitingexpansionofimmatureuNKcellswithinMLAp,whiletheTSLPsignalingpathwayisusedinDBtosustainIFN‐γproductionfromasubsetofmatureuNKcells.Regionalized,dynamicexpressionoftheadditionallymphoidorganstromalmarkersgp38/podoplaninandER‐TR7,butnotCD157,wereseenbyimmunohistochemistryinimplantationsites,andDBandMLApcontainedtranscriptsforAire,atolerance‐promotingfactor.TheseobservationssuggestthatCD127+NKlineageprogenitorsarenotpresentintheearlypostimplantationperiodofmouseuterusandthatdecidualizedendometrialstromahaskeyimmunoregulatoryproperties...
