【 摘 要 】
ThecausativemicroorganismsdictatethetypeofMDSCgeneratedinsepsispatients,andalargeproportionofPMN‐MDSCsingram‐positivesepsisincludesimmunosuppressivemyeloidblasts.MDSCsconstituteaheterogeneouspopulationofimmaturemyeloidcellsthatpotentlysuppressimmuneresponses.Theywereidentifiedoriginallyincancerpatientsandhavesincebeenreportedtooccuralsoinchronicinflammation,autoimmunity,andevenbacterialinfections.HumanMDSCsarecommonlydividedintoMo‐MDSCsandgranulocytic(PMN‐MDSCs)subtypes.TowhatextentthebonafidecancerMDSCsarerepresentativeoftheproposedMDSCsfoundinotherdiseasesisnotwellknown.PMN‐MDSCshavebeenfoundpreviouslytobeenrichedamongLDGsindensitygradient‐centrifugedblood.Inthisstudy,weanalyzedpotentialMDSCsinsepsispatientswithdifferentcausativemicroorganisms,usingtotalperipheralbloodcomparedwithdensitygradient‐centrifugedblood.WefoundahighfrequencyoftypicalCD14+HLA‐DRlowMo‐MDSCsinallsepsispatients,whereasthetypicalPMN‐MDSCs,aswellasaprominentCD14lowPMN‐MDSC‐likepopulation,appearedpreferentiallyingram‐positivecases.TheCD14lowPMN‐MDSCvariantwasdemonstratedtosuppressTcellproliferationinvitroviaaROS‐dependentmechanism,todisplayanincreasedIL‐10:TNF‐αratio,andtopresentwithsignsofimmaturity:blastmorphologyandlowcytokinelevels.WeconcludethataspectrumofcellswithMDSCfeaturesisenrichedinsepsisandthatthemicrobialoriginofsepsiscontributestothesubstantialinterindividualpatientvariationintheMDSCpattern...
