HumanpDCsrepresentararepopulationofcirculatingcellscharacterizedbyarapidandmassiveTLR‐dependentsecretionoftypeIIFNinresponsetopathogenicagentsordangersignals.Throughtheircapacitytobringtogetherinnateandadaptiveimmunityandtosecretesolublefactorscontrollingcancerdevelopment,thesecellscouldrepresentimportantactorsinantitumorimmunity.However,accumulatingevidencesuggeststhatpDCsrecruitedtothetumormicroenvironmentoftendisplayanonactivatedstateandareassociatedwiththedevelopmentandmaintenanceofimmunosuppression.Here,wepresentanoverviewofneoplasticlesionsassociatedwithaninfiltrationofimmunosuppressive/tolerogenicpDC.Moreover,astheproperresponseofpDCagainstcancerdependsonacriticalbalancebetweenimmune‐activatingandimmune‐suppressingmechanisms,wesummarizecurrentknowledgeaboutthemolecularpathwaysdevelopedbytumorstopreventantitumoralpDCimmuneresponses.AbetterunderstandingofthemechanismsregulatingpDCfunctionintumorscouldaidinthedevelopmentofnewtherapies.Indeed,effectivecancervaccinesortherapiescouldcombineimmunoactivatingstrategies(i.e.,TLRagonists)witheliminationofimmune‐suppressingmechanisms,leadingtopDCreprogrammingandthus,allowingtumorrejectioninaclinicalsetting...
