Bacterialandviralinfectionscausehighratesofmorbidityandmortalityinprematurenewborns.Inthesettingofviralinfection,pDCsplayakeyroleasstrongproducersofIFN‐αuponTLR9activation.WeanalyzedpDCfrequency,phenotype,morphology,andfunctioninCBofpretermandtermnewbornsincomparisonwithadults.WhereasallagegroupsshowsimilarpDCnumbers,BDCA‐2,CD123,andTLR9levels,theexpressionofBDCA‐4andcapacitytoproduceIFN‐αuponTLR9challengeweredecreasedsignificantlyinpretermneonates.Furthermore,weshowbymeansofelectronmicroscopythatpDCsfrompretermnewbornsexhibitadistinct,“immature”morphology.Takentogether,thesefindingssuggestdecreasedfunctionalityofpDCsintheprematurenewborn.ThereducedcapacitytoproduceIFN‐αislikelytorendersuchinfantsmoresusceptibletoviralinfections...
