WehavedemonstratedpreviouslythatBM‐derivedDCscanpreventdiabetesdevelopmentandhaltprogressionofinsulitisinNODmice,themousemodeloftype1diabetes.TheDCpopulationthatwasmosteffectiveinthistherapyhadamaturephenotype,expressedhighlevelsofcostimulatorymolecules,andsecretedlowlevelsofIL‐12p70.TheprotectiveDCtherapyinducedTregandTh2cellsinvitroandinvivo.MicroarrayanalysisoftherapeuticandnontherapeuticDCpopulationsrevealeddifferencesintheexpressionofOX40L,CD200,Ym‐1,CCL2,andCCL5,whichcouldplayimportantrolesintheobservedDC‐mediatedtherapy.TheuniquepatternofcostimulatorymoleculesandchemokinesexpressedbythetherapeuticDCswasconfirmedbyflowcytometryandELISA.Usinganovelcell‐labelingand19FNMR,weobservedthatthechemokinessecretedbythetherapeuticDCsalteredthemigrationofdiabetogenicTh1cellsinvivoandattractedTh2cells.TheseresultssuggestthatthetherapeuticfunctionofDCsismediatedbyacombinationofcostimulatoryandchemokinepropertiesthatresultsintheattractionofdiabetogenicTh1andtheinductionofTh2and/orTregdifferentiation...
