TLRsmediaterecognitionofawiderangeofmicrobialproducts,includingLPS,lipoproteins,flagellin,andbacterialDNA,andsignalingthroughTLRsleadstotheproductionofinflammatorymediators.InadditiontoTLRs,manyothersurfacereceptorshavebeenproposedtoparticipateininnateimmunityandmicrobialrecognition,andsignalingthroughsomeofthese,forexample,C‐typelectins,islikelytocooperatewithTLRsignalingindefininginflammatoryresponses.Inthepresentstudy,weexaminedtheimportanceoftheECDandintracellularTIRdomainofboTLR2andhuTLR2toinduceaspecies‐specificresponsebycreatingachimericTLR2protein.OurresultsindicatethatthestrengthoftheresponsetoanyTLR2ligandtestedwasdependentontheextracellular,solenoidstructure,butnottheintracellularTIRdomain.Furthermore,weexaminedwhethertherecognitionoftwoPAMPsbyDectin‐1,aCLR,dependsontheinteractionwithTLR2fromthesamespecies.TLR2expressionseemedtoaffecttheDectin‐1‐dependentproductionofCXCL8toβ‐glucancontainingzymosanaswellasListeriamonocytogenes.Furthermore,theinteractionofDectin‐1withTLR2seemedtorequirethatbothreceptorsarefromthesamespecies.OurdatademonstratethatthedifferencesintheTLR2responseseenbetweenthebovineandhumansystemdependontheECDofTLR2andthatcollaborativerecognitionofdistinctmicrobialcomponentsbydifferentclassesofinnate‐immunereceptorsiscrucialinorchestratinginflammatoryresponses...
