TLRsplayacriticalroleinthedetectionofmicrobesandendogenous“alarmins”toinitiatehostdefense,yettheycanalsocontributetothedevelopmentandprogressionofinflammatoryandautoimmunediseases.Toavoidpathogenicinflammation,TLRsignalingissubjecttomultilayerregulatorycontrolmechanisms,includingcooperationwithcoreceptors,post‐translationalmodifications,cleavage,cellulartrafficking,andinteractionswithnegativeregulators.Nucleicacid‐sensingTLRsareparticularlyinterestinginthisregard,astheycanbothrecognizehost‐derivedstructuresandrequireinternalizationoftheirligandasaresultofintracellularsequestrationofthenucleicacid‐sensingTLRs.ThisreviewsummarizestheregulatorymechanismsofTLRs,includingregulationoftheiraccesstoligands,receptorfolding,intracellulartrafficking,andpost‐translationalmodifications,aswellashowalteredcontrolmechanismcouldcontributetoinflammatoryandautoimmunedisorders...
