Inhumans,mutationsinATGLleadtoTGaccumulationinLDsofmosttissuesandcells,includingperipheralbloodleukocytes.ThispathologicconditioniscalledJordans’anomaly,inwhichfunctionalconsequenceshavenotbeeninvestigated.Inthepresentstudy,wetestedthehypothesisthatATGLplaysaroleinleukocyteLDmetabolismandimmunecellfunction.Similartohumanswithloss‐of‐functionmutationsinATGL,wefoundthatglobalandmyeloid‐specificAtgl−/−miceexhibitJordans’anomalywithincreasedabundanceofintracellularTG‐richLDsinneutrophilgranulocytes.Inamodelofinflammatoryperitonitis,lipidaccumulationwasalsoobservedinmonocytesandmacrophagesbutnotineosinophilsorlymphocytes.NeutrophilsfromAtgl−/−miceshowedenhancedimmuneresponsesinvitro,whichweremoreprominentincellsfromglobalcomparedwithmyeloid‐specificAtgl−/−mice.Mechanistically,ATGL−/−aswellaspharmacologicalinhibitionofATGLledtoanimpairedreleaseoflipidmediatorsfromneutrophils.ThesefindingsdemonstratethatthereleaseoflipidmediatorsisdependentontheliberationofprecursormoleculesfromtheTG‐richpoolofLDsbyATGL.OurdataprovidemechanisticinsightsintoJordans’anomalyinneutrophilsandsuggestthatATGLisapotentregulatorofimmunecellfunctionandinflammatorydiseases...
