Onceviewedassimplyantibacterialeffectorcellspackedwithantimicrobials,neutrophilsarenowincreasinglyappreciatedfortheirregulatoryrolesinimmunityandinflammation.Thehomeostaticregulationofneutrophilsisthuscrucialforoptimaloperationoftheimmunesystem.Anattractivemodeltounderstandmechanisticallytheroleofneutrophilsisperiodontitis,anoralinflammatorydiseasethatisparticularlysensitivetoneutrophilalterationsinnumbersorfunction.Therecruitmentandproperactivationofneutrophilsarelargelydependentonleukocyteintegrinsandcomplement.Thisreviewdiscusseshowtheseprocessesareaffectedbyhostgeneticormicrobialfactorsleadingtothedevelopmentofperiodontitis.Forinstance,bothhypo‐andhyper‐recruitmentofneutrophilsasaresultofdeficienciesintheexpressionofβ2integrinsortheirnegativeregulators,respectively,causesunwarrantedIL‐17‐dependentinflammatoryboneloss.Moreover,microbialhijackingofC5aR(CD88)signalinginneutrophilsimpairstheirantimicrobialfunctionwhilepromotingdestructiveinflammatoryresponses.Thesestudiesnotonlysupporttheconceptthatneutrophilhomeostasisiskeytoperiodontalhealthbutalsorevealpromising,newtherapeutictargetsasdiscussedinthereview...
