【 摘 要 】
Generalizedaggressiveperiodontitis(GAgP)isaninflammatorydiseaseofhostresponsetobacterialchallenge.Toexploretheroleofplateletsinhost–microbialinteractionsinpatientswithperiodontitis,124patientswithGAgPand57healthysubjectswereenrolled.Reliableindicatorsofsubclinicalplateletfunctionalstatus,plateletcount(PLT),plateletlargecellratio(PLCR),andmeanplateletvolume(MPV),weresignificantlylowerintheGAgPgroupthaninthecontrolgroupandwerenegativelycorrelatedwithclinicalperiodontalparameters.Thelevelsofimportantcytosolicproteininneutrophils,calprotectin(S100A8/A9)inplasma,andgingivalcrevicularfluid(GCF)weresignificantlyhigherinpatientswithGAgPcomparedwithhealthysubjects.Moreover,theGCFcalprotectinlevelwasnegativelycorrelatedwithPLCRandMPVvalues.Toexplorethepossiblemechanismsofchangesinplateletindicesinperiodontitis,flowcytometryanalysiswasperformed,andpatientswithGAgPwerefoundtohaveahigherstatusofplateletactivationcomparedwithhealthycontrols.Porphyromonasgingivalis(P.gingivalis)andrecombinanthumanS100A8/A9(rhS100A8/A9)inducedplateletactivationandfacilitatedplatelet–leukocyteaggregateformationinwholebloodofhealthysubjects.InresponsetoP.gingivalisandrhS100A8/A9,plateletsfrompatientswithGAgPincreasedactivationandincreasedformationofplatelet–leukocyteaggregatescomparedwiththosefromhealthysubjects.PlateletaggregatesandplateletsattachedtoleukocyteswerefoundongingivaltissuesfrompatientswithGAgP,suggestingthatdecreasedplateletsizeandcountinthecirculationmightberelatedtoconsumptionoflarge,activatedplateletsatinflamedgingiva.Plateletsmayhaveapreviouslyunrecognizedroleinhostresponsetoperiodontalinfection...
